Description

Zilretta (triamcinolone acetonide) is an extended-release intra-articular corticosteroid injection approved for the management of osteoarthritis-related knee pain. It is formulated with microsphere technology, allowing for sustained drug release and prolonged therapeutic effects. The corticosteroid reduces inflammation by inhibiting phospholipase A2 and suppressing the release of inflammatory mediators such as prostaglandins and leukotrienes.

Each kit includes a single-dose vial containing 32 mg of triamcinolone acetonide microsphere powder, a vial of diluent (5 mL), and a sterile vial adapter for reconstitution. The microsphere formulation ensures gradual drug release over an extended period, reducing the need for repeated injections. Zilretta should be administered by intra-articular injection only and is not intended for repeat use.

Common adverse effects include joint pain, headache, and injection site reactions. Serious complications include joint infection, hypersensitivity reactions, and systemic corticosteroid effects such as adrenal suppression or hyperglycemia. Patients should be monitored for infection and systemic corticosteroid exposure, particularly those with diabetes or immunosuppression.
