Description
Tacrolimus 0.1% topical solution is indicated for the treatment of moderate to severe atopic dermatitis in patients who are unresponsive to or intolerant of conventional therapies. As a calcineurin inhibitor, tacrolimus suppresses T-cell activation and cytokine release, reducing inflammation and pruritus in affected skin areas. It is applied as a thin layer to the affected regions and is suitable for short-term or intermittent long-term use.
The formulation carries a black box warning due to the potential increased risk of lymphoma and skin malignancies. Patients may also experience localized adverse effects such as a burning sensation, pruritus, or erythema at the site of application. Use should be limited to non-occluded skin, avoiding application under occlusive dressings or to mucous membranes.
Concurrent use with other immunosuppressive agents should be avoided due to the risk of additive immunosuppression and potential systemic toxicity. Hypersensitivity to tacrolimus or any of the inactive ingredients in the formulation is a contraindication. Off-label uses include management of conditions such as vitiligo and psoriasis.
